What is Buffy Coat?
A buffy coat is the thin, yellowish-white layer of leukocytes (white blood cells) and platelets (thrombocytes) that forms between the red blood cells and plasma after centrifugation of anticoagulated blood. Furthermore, this concentrated layer is a valuable resource for various applications, including DNA extraction, because it contains nucleated white blood cells.
In addition, in the blood banking industry, blood banks often collect buffy coats from whole blood donations to produce platelet concentrates for transfusion, especially for patients with cancer, trauma, or hematological diseases.
Buffy Coat is processed from patients’ whole blood samples collected in EDTA anticoagulant tubes using centrifugation. After the centrifugation process, a layer of clear fluid, the plasma, comes on top; meanwhile, a layer of red fluid (Erythrocytes), which contains most of the red blood cells, stays at the bottom.
Additionally, a very thin white layer in between is the buffy coat. Each of the buffy coats is less than 1% of the total blood volume. Notably, the buffy coat is so-called because it is usually buff in hue.
- Top Layer – Plasma: 55% of total Blood Volume
- Middle Layer – Buffy Coat: Leukocytes and Platelets <1% of total Blood Volume
- Bottom Layer – Erythrocytes: 45% of total Blood Volume
Furthermore, the buffy coat consists of white blood cells and platelets, and it is a concentration of leukocyte suspension. Importantly, the buffy coat is used to extract DNA from the blood of mammals, since mammalian red blood cells are anucleate and do not contain DNA.
Consequently, this offers an option for purifying large amounts of gDNA (genomic DNA) from relatively small sample sizes. Additionally, buffy coat preparation is often the first step towards further processing for T-cell depletion, tumor cell purging, or cryopreservation for both marrow and blood hematopoietic progenitor cell (HPC) collections.
Preparation
Buffy coat is derived by processing whole blood using the following procedure:
- First, collect anticoagulated blood: A technician collects blood into tubes containing an anticoagulant.
- Next, centrifuge the blood: The technician centrifuges the blood sample at high speed.
- Then, separate the layers: This process separates the blood into three distinct layers:
- Specifically, Plasma: (top layer)
- Followed by, Buffy coat: (middle, thin layer of white blood cells and platelets)
- And finally, Erythrocytes: or red blood cells (bottom layer)
Components of a Buffy Coat
The buffy coat contains a mix of immune cells, including:
- Firstly, Granulocytes: Neutrophils, basophils, and eosinophils.
- Additionally, Lymphocytes: B cells, T cells, and natural killer cells.
- Moreover, Monocytes: White blood cells that can develop into macrophages or dendritic cells.
- Finally, Platelets (Thrombocytes): Small cell fragments important for blood clotting.
Applications
