Buffy coat is processed from patients whole blood samples collected in EDTA anticoagulant tube using centrifugation. After centrifugation process a layer of clear fluid, the plasma comes on top, and a layer of red fluid (Erythrocytes) containing most of the red blood cells stays at the bottom, and a very thin white layer in between is the Buffy coat. Each of the Buffy coat is less than 1% of the total blood volume. The Buffy coat is so-called because it is usually buff in hue.
Top Layer- Plasma: 55% of total Blood Volume
Middle Layer- Buffy Coat: Leukocytes and Platelets <1% of total Blood Volume
Bottom Layer- Erythrocytes: 45% of total Blood Volume
Buffy coat consists of white blood cells, platelets and is a concentration of leukocyte suspension. The Buffy coat is used to extract DNA from the blood of mammals, since mammalian red blood cells are anucleate and do not contain DNA. This offers an option for purifying large amounts of gDNA (genomic DNA) from relatively small sample sizes. Buffy coat preparation is often the first step towards additional processing for T-cell depletion, tumor cell purging or cryopreservation for both marrow and blood Hematopoietic progenitor cells (HPC) collections.
