Serum Plasma Biofluid Processing Protocol

     SERUM AND PLASMA PROCESSING PROTOCOL

Sample Collection and Processing Overview: Standard protocol for serum and plasma collection will be as follows:

1).Confirm and make sure the signed and dated Bay Biosciences LLC., informed consent form (ICF) was obtained from the patient.

2). Draw whole blood in one 10 mL serum separator collection tube (tiger top).

3). Draw whole blood in one 10 mL EDTA or K2EDTA blood collection tube (lavender top), and invert 6 to 8 times at room temperature.

Steps 4-11 must be carried out within 30-60 minutes of collection to prevent biochemical changes.

4). Centrifuge the tubes for 20 minutes at 1300 x g (approximately 3250 RPM)  to separate the serum/plasma from the cells.

Aliquoting

5). Pipette a minimum of 1 mL serum from the tiger top (serum separator) tube into each pre-labelled aliquot vial. Depending on the hematocrit of the patient, one 10 mL tube of blood yields 3 to 5 aliquots of serum in volumes of 1 mL per vials. The average yield from 10 mL blood collection is total of 4 mL of serum.

6). Record the pre-labeled cryovial donor identification number (DIN) on the procurement log and label as serum.

7). Pipette a minimum of 1mL plasma from the lavender top (EDTA) tube into each pre-labeled aliquot vial. Depending on the hematocrit of the patient, one 10 mL tube of blood yields 3 to 5 aliquots of plasma in volumes of 1 mL per vials. The average yield from 10 mL blood collection is a total of 4 mL of plasma.

8). Record the pre-labeled cryovial donor identification number (DIN) on the procurement log and label as plasma.

9). Pipette a minimum of 1 mL of the peripheral blood cells that remain in the bottom of the lavender top (EDTA) tube into a pre-labeled cryovial.

10). Record the pre-labeled cryovial donor identification number on the procurement log and label as cells.

11). Snap-freeze aliquots and store frozen at -20° C or lower. If liquid nitrogen is unavailable, place samples directly into -80° C freezer unit. It is important to avoid freeze-thaw cycles.
